Video Tutorials on Ultrathin sectioning
Posted on 5 March 2023
This video goes through an entire workflow of regular ultrathin sectioning using a diamond knife, starting from a trimmed resin block. It is useful to those who are starting to learn ultramicrotomy. As it is designed to train inexperienced users, it shows a section collection technique that is relatively easy and ‘trouble-free’ (collecting sections with a loop, rather than by capturing with a grid from below or above the sections). Although it is not designed as a substitute for face-to-face training (it doesn’t cover all tricks), it has supported training very successfully in our facility. It covers basic ultrathin sectioning, but not serial sectioning. Block trimming prior to ultramicrotomy is covered in other videos found on the Oxford Brookes Centre for Bioimaging website.
